Analysis of Drugs: Confirmation and Quantification of Methadone by Master GC coupled with Master TOF-MS Plus
Methadone is a prescription opioid used primarily as pain relief, treatment of opioid abstinence syndromes and treatment of heroin users. The selection of methadone for use in substance abuse treatment programs is based in part on its relatively long duration of action in suppressing withdrawal compared with other opiates such as morphine and meperidine.
Methadone is metabolized by demethylation and cyclation to 2-ethylidene-1,5-dimethyl-3,3- diphenylpyrrolidine (EDDP), in turn converted to 2-ethyl-5-methyl-3,3-diphenylpyrrolidine (EMDP).
The Cyclic structure of EDDP and EMDP makes them very difficult to detect using immunoassays targeted to the parent compound methadone.
Master GC coupled with Master TOF MS Plus allows the identification and quantification of the drug and its secondary metabolite, under the concentration limit (United States frequently used Cut Off: 300ng/ mL).
The system allows the analysis of Standard and Real Autopsy Samples in complex Matrix (Serum) with high sensitivity and linearity, with a Total Analysis Time that is less than 7 minutes.
